DBH
dopamine beta-hydroxylase
Gene summary
A protein-coding gene on chromosome 9 (neurotransmission). The protein encoded by this gene is an oxidoreductase belonging to the copper type II, ascorbate-dependent monooxygenase family. The encoded protein, expressed in neuroscretory vesicles and chromaffin granules of the adrenal medulla, catalyzes the conversion of dopamine to norepinephrine, which functions as both a hormone and as the main neurotransmitter of the sympathetic nervous system.

Protein reference (UniProt)
- Protein
Dopamine beta-hydroxylase
- Function
Catalyzes the hydroxylation of dopamine to noradrenaline (also known as norepinephrine), and is thus vital for regulation of these neurotransmitters.
- Pathway
Catecholamine biosynthesis; (R)-noradrenaline biosynthesis; (R)-noradrenaline from dopamine: step 1/1.
- Subcellular location
Cytoplasmic vesicle, secretory vesicle lumen; Cytoplasmic vesicle, secretory vesicle, chromaffin granule lumen; Secreted; Cytoplasmic vesicle, secretory vesicle membrane; Cytoplasmic vesicle, secretory vesicle, chromaffin granule membrane.
- Associated conditions
Orthostatic hypotension 1 (ORTHYP1).

Why its pathway is in the library
Monoamine, GABA, glutamate, and cholinergic systems are the direct pharmacologic targets of SSRIs/SNRIs (PSSD) and the downstream effectors of neurosteroid signaling (PFS).
Written for the whole neurotransmission family, not for DBH specifically.

Comment in a thread on emerging trends in PSSD genomes involving the DBH gene. Powers notes that several PSSD patients have volunteered for the temporary chemical-castration trial if his other PSSD approaches don't work. He says he's increasingly convinced of…

Post body unavailable. From the title, Powers announced he was beginning to see recurring trends in the genomes of his PSSD patients, singling out DBH — dopamine beta-hydroxylase, the enzyme converting dopamine to norepinephrine — as one such trend. This is…

Asked whether DHEA is still used alongside progesterone/pregnenolone for neurosteroid-type patients, Powers splits PSSD into androgenic-metabolite vs strictly-neurosteroid cases. Pregnenolone: yes for strictly-neurosteroid PSSD — it backfills the DHEA…
