GPX4
glutathione peroxidase 4
Pathway candidate — not linked to PFS or PSSD by any record here
Gene summary
A protein-coding gene on chromosome 19 (mitochondrial function and energy). The protein encoded by this gene belongs to the glutathione peroxidase family, members of which catalyze the reduction of hydrogen peroxide, organic hydroperoxides and lipid hydroperoxides, and thereby protect cells against oxidative damage. Several isozymes of this gene family exist in vertebrates, which vary in cellular location and substrate specificity.

Phospholipid hydroperoxide glutathione peroxidase GPX4
- Function (excerpt)
Essential antioxidant peroxidase that directly reduces phospholipid hydroperoxide even if they are incorporated in membranes and lipoproteins. Can also reduce cholesterol hydroperoxide and thymine hydroperoxide (By similarity). Plays a key role in protecting cells from oxidative damage by preventing membrane lipid peroxidation. Required to prevent cells from ferroptosis, a non-apoptotic cell death resulting from an iron-dependent accumulation of lipid reactive oxygen species. The presence of selenocysteine (Sec) versus Cys at the active site is essential for life: it provides resistance to overoxidation and prevents cells against ferroptosis (By similarity). The presence of Sec at the active site is also essential for the survival of a specific type of parvalbumin-positive interneurons, thereby preventing against fatal epileptic seizures (By similarity). May be required to protect cells from the toxicity of ingested lipid hydroperoxides (By similarity).
- Subcellular location
Mitochondrion; Cytoplasm.
- Tissue specificity
Present primarily in testis. Expressed in platelets (at protein level).
- Associated conditions
Spondylometaphyseal dysplasia, Sedaghatian type (SMDS).

Why its pathway is in the library
Mitochondrial dysfunction and oxidative stress are reported in PFS/PSSD patient work; energy failure plausibly contributes to fatigue, cognitive, and sexual symptoms.
Written for the whole mitochondria & energy family, not for GPX4 specifically.
Mentioned in 0 corpus records
No corpus record names GPX4 directly yet. It is in the library because it sits in a pathway the corpus tracks (Mitochondria & energy).
